These samples serve as baseline and matched control material for immunology, haematology, genomics, and pharmacology research.
Why Whole Blood Rather Than an Isolated Fraction
Whole blood is the only blood specimen that retains every cellular and soluble component in its native proportion, because nothing has been removed. Every downstream fraction — plasma, serum, PBMC, isolated subsets — is produced by a processing step that changes what is present.
- Density gradient PBMC preparation discards granulocytes; whole blood retains neutrophils, eosinophils, and basophils, which is essential for any study where granulocyte biology is part of the question.
- Cells remain in autologous plasma, so whole blood supports stimulation assays in which the physiological milieu — complement, immunoglobulin, acute phase proteins, plasma protein binding — is preserved rather than replaced with culture medium.
- Avoiding an isolation step avoids isolation-induced activation and marker shedding, which matters for accurate baseline immunophenotyping by flow cytometry.
- Whole blood is the required input for complete blood count and differential, and for haematology instrument and assay validation.
- It is the standard source for high-molecular-weight genomic DNA and, in RNA-stabilising tubes, for whole-blood transcriptomic profiling that captures the granulocyte-inclusive signature.
- Anticoagulant choice is not a formality: EDTA is conventional for cell counts and DNA extraction, heparin for functional culture and stimulation, citrate for coagulation work, and RNA-stabilising tubes for transcriptomics. Specifying the tube up front prevents an unusable specimen.
