What Is Human Serum?

Human serum is the clear, liquid fraction of blood that remains after the blood has clotted and the clot (including cells and clotting factors such as fibrinogen) has been removed. Serum contains proteins, electrolytes, antibodies, antigens, hormones, and metabolites, but — unlike plasma — it contains no fibrinogen or other clotting factors. This makes serum a preferred matrix for many biomarker, antibody, and diagnostic assays.

Serum vs. Plasma

The key difference is how the sample is processed: serum is collected after blood is allowed to clot (no anticoagulant), while plasma is collected with an anticoagulant (EDTA, heparin, or citrate) so the sample stays liquid and retains clotting factors. Serum is typically chosen for serology and many immunoassays; plasma is chosen when clotting factors or the fastest processing are required. Off-the-Clot vs. AB Serum

Off-the-clot serum is produced by allowing blood to clot naturally and separating the serum, preserving native analyte levels for research. Human AB serum is pooled serum from type-AB donors (lacking anti-A and anti-B antibodies), commonly used as a cell-culture supplement. Sanguine can supply single-donor off-the-clot serum as well as defined pooled human serum (pooled human sera) to match your protocol.

Normal Human Serum (NHS)

Normal human serum — also called healthy human serum or NHS — is serum from screened donors with no diagnosed condition relevant to your study. It is the standard control matrix and assay diluent for immunoassay development, and the baseline arm in any case-control design. Available as:

  • Single-donor normal human serum — preserves individual variation, and the right choice when you need donor-level demographic or genotype linkage
  • Pooled normal human serum — averages out individual variation for a consistent, reproducible matrix across a study
  • Demographically matched controls — age- and sex-matched to a disease cohort, which is usually the correct comparator rather than convenience controls

Pooled Human Serum Specifications

Pooled human sera are prepared to defined specifications so that results are reproducible across a programme:

  • Pool size — specified donor count per lot; larger pools give tighter analyte consistency
  • Blood type — AB pools for cell culture, or mixed/type-specific pools on request
  • Filtration — 0.2 µm sterile-filtered on request
  • Heat inactivation — available where complement activity must be removed
  • Lot reservation — reserve a single lot for the duration of a study to eliminate lot-to-lot variability as a variable
