LeukoCore Inflammatory Bowel Disease Leukopak biospecimens are collected by IRB-approved apheresis from donors with a clinically confirmed diagnosis of inflammatory bowel disease. Each leukopak yields large quantities of peripheral blood mononuclear cells, processed and shipped within 24 hours of collection, making it suited to high-input immune profiling, cell isolation, and drug discovery work in IBD.

IBD Immunopathology and Why a Leukopak

IBD is an immune-mediated inflammatory disease of the gastrointestinal tract, driven by a dysregulated mucosal immune response to the gut microbiota in a genetically susceptible host with a compromised epithelial barrier. It is not a classical autoantibody-mediated autoimmune disease:

• Two principal forms. Crohn’s disease can affect any part of the tract with transmural, patchy inflammation and granulomas; ulcerative colitis is a continuous, mucosal colonic inflammation extending proximally from the rectum
• Genetic susceptibility includes NOD2, ATG16L1, and IL23R, implicating bacterial sensing, autophagy, and the IL-23/Th17 axis in disease risk
• Cytokine pathways are the drug targets. TNF, IL-12/IL-23, IL-17, JAK-STAT signalling, and lymphocyte trafficking via the α4β7 integrin–MAdCAM-1 axis all underpin approved and investigational therapies
• Loss of response is a central clinical problem — primary non-response and secondary loss of response to anti-TNF therapy make pre-treatment donor material valuable for predictive biomarker work
• Why a leukopak: apheresis provides cell numbers far beyond a standard blood draw, enough for multi-parameter flow cytometry, magnetic or FACS isolation of T-cell, B-cell, monocyte and NK subsets, and repeat stimulation experiments from a single donor
• Fresh or cryopreserved formats support same-day functional assays or banked, batched study designs

Donor Stratification Available

• Diagnosis: Crohn’s disease or ulcerative colitis
• Disease location and extent, including ileal, colonic, ileocolonic, or perianal involvement
• Disease activity: active flare versus clinical remission
• Treatment status: treatment-naive, corticosteroids, immunomodulators, anti-TNF, anti-integrin, anti-IL-12/23, or JAK inhibitor therapy
• Surgical history, including prior resection or ostomy
• Age at diagnosis, disease duration, sex assigned at birth, and race/ethnicity
• Matched healthy controls available
