Inflammatory Bowel Disease PBMC products are sourced from IRB-consented donors clinically diagnosed with inflammatory bowel disease (IBD), processed within 24 hours of collection and cryopreserved in CryoStor CS10. These peripheral blood mononuclear cells support cellular immunology work on IBD-associated immune dysregulation.
IBD Immunology and Why PBMC
IBD is an immune-mediated inflammatory disease of the gastrointestinal tract, comprising Crohn’s disease and ulcerative colitis. It is driven by dysregulated mucosal immunity to the gut microbiota in genetically susceptible individuals rather than by a defined autoantibody.
- Barrier dysfunction and loss of tolerance to commensal microbiota allow inappropriate activation of mucosal T cells, with effector cytokine production sustaining chronic inflammation
- The TNF-alpha and IL-23/Th17 axes are the two best-validated therapeutic pathways, targeted respectively by anti-TNF and anti-IL-12/23 and anti-IL-23 biologics
- Lymphocyte trafficking to the gut, mediated in part by integrin alpha4beta7 and its interaction with MAdCAM-1, is a clinically validated target and is directly measurable on circulating cells
- Genome-wide association studies implicate many loci, with NOD2 variants a well-established contributor to Crohn’s disease susceptibility
- Crohn’s disease and ulcerative colitis differ in distribution, depth of inflammation and treatment response, so subtype selection matters
- PBMC support flow cytometric immunophenotyping including gut-homing marker expression, single-cell sequencing, ex vivo stimulation and functional cytokine assays
