Vasculitis PBMC for Immune-Mediated Vascular Inflammation Research

Vasculitis Human PBMC products are sourced from IRB-consented donors clinically diagnosed with vasculitis, processed within 24 hours of collection and cryopreserved in CryoStor CS10. These peripheral blood mononuclear cells support cellular immunology work on the immune mechanisms that drive blood-vessel inflammation.

Vasculitis Immunology and Why PBMC

Vasculitis is an umbrella term for inflammation of blood-vessel walls, and the underlying immune mechanism differs by subtype. Sanguine does not claim a single unifying autoantibody mechanism across the group, so subtype selection matters when designing a study.

  • ANCA-associated vasculitis, including granulomatosis with polyangiitis and microscopic polyangiitis, is autoimmune and associated with antibodies to proteinase 3 or myeloperoxidase, with neutrophil activation central to the pathology
  • Large-vessel vasculitis, including giant cell arteritis and Takayasu arteritis, is immune-mediated and granulomatous, with T cell and macrophage-driven arterial wall inflammation
  • IgA vasculitis is an immune-complex-mediated small-vessel disease, and behaves differently again
  • Across subtypes, circulating monocytes, T cell subsets and B cells carry disease- and treatment-related signatures, which is what makes viable PBMC the appropriate specimen
  • PBMC support flow cytometric immunophenotyping, single-cell and bulk RNA sequencing, ex vivo stimulation, and functional cytokine-release assays
  • Because many patients receive glucocorticoids, rituximab or other immunosuppression, treatment status is a critical covariate and is captured in the donor record

  • Subtype: ANCA-associated, large-vessel (giant cell arteritis or Takayasu), or immune-complex vasculitis
  • ANCA serology where available, including PR3-ANCA and MPO-ANCA specificity
  • Disease state: active disease versus remission
  • Treatment status: treatment-naive, on glucocorticoids, on conventional immunosuppression, or on B cell depleting therapy
  • Documented organ involvement, such as renal, pulmonary, or cutaneous
  • Matched healthy controls available
