Healthy CD19+ B Cells for Immunology and Antibody Discovery Research

Healthy CD19 B Cells are purified from the peripheral blood of healthy, IRB-consented, clinically screened adult donors and cryopreserved within 24 hours of collection. B lymphocytes are the effectors of humoral immunity, and these primary human cells serve as the baseline reference material for studying antibody production, B cell receptor (BCR) signalling, and immune regulation. Disease-state CD19+ B cells are also available for comparative work.

B Cell Biology and Isolation

  • CD19 is the canonical pan-B-cell marker, expressed from the pro-B stage through to activated B cells and most plasmablasts, and lost on fully differentiated plasma cells. It is also the most clinically established target antigen in B-cell-directed cell therapy.
  • Cells are isolated from apheresis or peripheral blood mononuclear cell starting material by immunomagnetic selection. Negative selection leaves the B cell surface untouched and is preferred where downstream receptor engagement or activation state matters; positive selection is available where maximum purity is the priority.
  • The isolated fraction contains the physiological spread of circulating B cell subsets, distinguishable by CD27, IgD, CD24, and CD38 expression: transitional and naive B cells, unswitched and class-switched memory B cells, and a small plasmablast population.
  • CD19 is physically and functionally coupled to the BCR complex, where it acts with CD21 and CD81 as a co-receptor that lowers the threshold for antigen-driven activation – making these cells directly suited to signalling studies.
  • Primary human B cells are short-lived in culture without support, so activation systems supplying CD40 ligand, IL-4, IL-21, or TLR9 stimulation are typically used for proliferation, class switching, and differentiation assays.
  • Cryopreservation in CryoStor CS10 with processing inside 24 hours of collection preserves viability and function for thaw-and-use assay formats.
