Our Transthyretin Amyloidosis (ATTR) Serum is sourced from IRB-consented donors clinically diagnosed with ATTR amyloidosis, a progressive disease in which misfolded transthyretin (TTR) protein aggregates into amyloid fibrils that deposit in peripheral nerves and the heart. Each sample is processed and frozen within 24 hours of collection to preserve circulating TTR, cardiac biomarkers, and other amyloid-relevant proteins.
ATTR Biology and Why Serum
- ATTR amyloidosis arises from destabilized TTR tetramers – either due to inherited point mutations (hereditary/variant ATTR, ATTRv) or age-related misfolding of wild-type TTR (ATTRwt) – that dissociate into monomers prone to aggregation and fibril formation.
- Amyloid deposition in peripheral and autonomic nerves produces a length-dependent polyneuropathy, while cardiac deposition causes restrictive cardiomyopathy with progressive heart failure and conduction abnormalities.
- Serum is widely used for TTR concentration and stability immunoassays and is the standard matrix for many clinical cardiac biomarker platforms (NT-proBNP, high-sensitivity troponin) that track amyloid cardiomyopathy severity.
- Approved and investigational therapeutics target distinct mechanisms: TTR tetramer stabilizers (e.g. tafamidis), TTR gene-silencing therapies (patisiran, inotersen, vutrisiran), and CRISPR-based knockdown approaches, all of which rely on circulating TTR level and stability measurements for pharmacodynamic monitoring.
- Genotype (e.g. Val142Ile, Val50Met) and phenotype (predominantly neuropathic, cardiac, or mixed) are key stratification variables for both natural history and treatment-response research.
