What are PBMCs?

Peripheral blood mononuclear cells (PBMCs) are the blood cells with a single, round nucleus: lymphocytes (T cells, B cells and NK cells) and monocytes, with a small fraction of dendritic cells. They are isolated from whole blood, buffy coat or a leukopak by density-gradient centrifugation, which separates them from red blood cells, granulocytes and plasma. Because they hold the main circulating immune-cell populations, PBMCs are used for immune-cell phenotyping, cellular functional assays, and single-cell and bulk transcriptomics.
