Atopic Dermatitis PBMC for Type 2 Immunity and Skin-Homing T Cell Research

Atopic Dermatitis PBMC products are sourced from IRB-consented donors clinically diagnosed with atopic dermatitis and processed under stringent conditions. These cryopreserved peripheral blood mononuclear cells enable functional study of the type 2-skewed immune response that drives atopic dermatitis — an allergic inflammatory skin disease in which barrier dysfunction and Th2 cytokine signalling reinforce one another, rather than an autoantibody-mediated condition.

Why PBMCs for Atopic Dermatitis Research

  • Circulating Th2 cells are the cellular source of IL-4, IL-13 and IL-31 in atopic dermatitis. PBMCs allow these populations to be phenotyped directly and, on ex vivo stimulation, to be measured for actual cytokine output rather than inferred from soluble levels.
  • Skin-homing T cells expressing cutaneous lymphocyte antigen (CLA) and CCR4 — the receptor for the atopic dermatitis-associated chemokine CCL17/TARC — circulate in blood and can be isolated and characterised from PBMCs without needing a biopsy.
  • Dupilumab blocks the shared IL-4 receptor alpha chain, so IL-4Ralpha surface expression and downstream STAT6 phosphorylation in PBMCs constitute a genuine target-engagement readout for the anti-type 2 biologic class.
  • JAK inhibitors used in atopic dermatitis act on JAK-STAT signalling common to IL-4, IL-13 and IL-31 receptors, making phospho-flow and signalling assays on patient PBMCs directly relevant to that mechanism.
  • Regulatory T cell frequency and function, and the balance between effector and regulatory compartments, are studied in atopic dermatitis and require intact viable cells.
  • Basophils and innate lymphoid cell populations relevant to type 2 immunity are present in the mononuclear fraction, supporting activation testing and innate type 2 biology in the same donor sample.
