Atopic Dermatitis Plasma for Type 2 Inflammation and Skin Barrier Research
Atopic Dermatitis Plasma is collected from IRB-consented donors clinically diagnosed with atopic dermatitis, a chronic form of eczema, and processed within one day of collection to preserve cytokines, chemokines, proteins, and metabolites. Atopic dermatitis is an allergic, type 2-skewed inflammatory disease in which epidermal barrier dysfunction and a Th2-dominated immune response reinforce each other; it is not an autoantibody-driven condition. Plasma from these donors supports research into circulating type 2 mediators, barrier biology, and treatment response.
Why Plasma for Atopic Dermatitis Research
- The core cytokines of atopic dermatitis — IL-4, IL-13, IL-5 and IL-31 — are the direct targets of dupilumab, tralokinumab, nemolizumab and the JAK inhibitor class. Plasma is the accessible matrix for measuring the soluble output of that axis and for pharmacodynamic work.
- CCL17 (TARC) is among the best-replicated circulating correlates of atopic dermatitis severity, and CCL22, periostin and TSLP-associated markers are studied alongside it. All are measurable in plasma.
- Barrier dysfunction is central to the disease. Filaggrin loss-of-function variants are the strongest established genetic risk factor, and plasma from genotyped donors allows soluble inflammatory readouts to be interpreted against barrier genotype.
- Total IgE and eosinophil-associated mediators reflect the type 2 skew and the atopic comorbidity burden — asthma, allergic rhinitis and food allergy — that frequently accompanies atopic dermatitis.
- Plasma avoids the ex vivo platelet degranulation that occurs during clotting, which matters when measuring platelet-stored chemokines such as CXCL4 and CCL5 that would otherwise be artefactually elevated in serum.
- Plasma drawn alongside a skin punch biopsy or PBMCs from the same donor lets circulating mediators be tied to lesional tissue and to the responsible cell populations.
