Healthy Pediatric PBMC for Immune Development and Control-Group Research

Healthy Pediatric PBMC samples are collected from screened healthy pediatric donors under IRB-approved protocols with guardian authorization, and processed under controlled conditions. These cryopreserved peripheral blood mononuclear cells provide baseline immune-cell profiles for studies of immune development, pediatric disease, and vaccine response.

Why Age-Matched Pediatric PBMC Matter

These are healthy-donor cells, so there is no disease mechanism to characterise — the scientific value lies in immune ontogeny. The paediatric immune system is not a smaller version of the adult one, and using adult PBMC as controls for a paediatric cohort introduces a systematic age artefact into every comparison.

  • Naive-dominant T cell repertoire — young children have a high proportion of naive T cells, with memory subsets accumulating progressively through childhood as antigen exposure builds
  • Active thymic output — thymic function is greatest in early life and declines with age, so recent thymic emigrant populations are present at frequencies not seen in adult donors
  • Age-dependent reference ranges — absolute lymphocyte counts and lymphocyte subset proportions differ substantially by age band, which is why paediatric normal ranges are defined separately from adult ones
  • Maturing B cell and NK compartments — immunoglobulin class-switched memory B cells and terminally differentiated NK subsets expand with age and antigen experience
  • Vaccine and infection response ontogeny — response magnitude and quality to vaccination differ by age, making age-matched paediatric baselines essential for vaccine immunology
  • Correct comparator design — for any paediatric disease cohort, age- and sex-matched healthy paediatric PBMC are the appropriate control arm rather than convenience adult controls
