Multiple Sclerosis CD56+ NK Cells are isolated from the peripheral blood of IRB-consented adult donors with a clinically confirmed diagnosis of multiple sclerosis and cryopreserved in CryoStor® CS10 within 24 hours of collection. These CD56+ CD3− natural killer cells provide a purified innate lymphocyte population for investigating immune regulation and cytotoxicity in autoimmune demyelinating disease.

NK Cell Biology in Multiple Sclerosis

Multiple sclerosis is an autoimmune demyelinating disease of the central nervous system in which autoreactive lymphocytes cross the blood-brain barrier and drive inflammatory injury to myelin and axons. NK cells occupy an unusual position in that process: rather than acting only as effectors, they appear to help restrain the autoreactive adaptive response, which is why they are a target of interest rather than simply a bystander population.

  • Human NK cells divide into a cytotoxic CD56dim majority and an immunoregulatory CD56bright subset; the CD56bright compartment is the one most consistently implicated in multiple sclerosis.
  • CD56bright NK cells can suppress activated autologous T cells, providing a mechanistic link between innate lymphocyte function and control of autoreactive T cell expansion.
  • Expansion of the CD56bright subset is an established pharmacodynamic effect of certain multiple sclerosis therapeutics, making NK phenotype a clinically anchored readout rather than a purely exploratory one.
  • NK activating and inhibitory receptor repertoires, including NKG2A, NKG2D and the killer-cell immunoglobulin-like receptors, shape cytotoxic potential and interaction with HLA ligands.
  • Disease-modifying therapies differ substantially in their effects on NK number and function, so treatment-annotated donors are essential to interpretable studies.
  • A purified CD56+ preparation permits direct cytotoxicity, degranulation and cytokine assays that cannot be resolved cleanly from a mixed mononuclear fraction.
