Thyroid Eye Disease Plasma for Autoimmune Orbital Disease Research

Thyroid Eye Disease Plasma is collected from IRB-consented donors clinically diagnosed with thyroid eye disease (TED) and processed within one day of collection to preserve cytokines, autoantibodies, proteins and metabolites. Plasma is acellular, so this product is intended for soluble-analyte measurement rather than cell-based assays.

TED Autoimmunity and Why Plasma

Thyroid eye disease is an autoimmune orbital condition, in the great majority of cases occurring in association with Graves’ disease. The orbit, not the thyroid, is the site of the pathology.

  • Stimulating autoantibodies against the thyrotropin receptor (TSHR), the same autoantibodies that cause Graves’ hyperthyroidism, are central to the disease and are detectable in circulation
  • Orbital fibroblasts express TSHR and IGF-1R, and functional crosstalk between these receptors is the mechanistic basis for IGF-1R-directed therapy in TED
  • Activated orbital fibroblasts secrete hyaluronan and a subset differentiate into adipocytes, expanding orbital soft tissue volume and producing proptosis and, in severe cases, optic neuropathy
  • An active inflammatory phase precedes a stable fibrotic phase, and clinical activity scoring is used to distinguish them; treatment strategy differs between the two
  • TSHR autoantibody levels, IGF-1 and inflammatory cytokines are all measurable in plasma, supporting non-invasive activity and treatment-response research without orbital biopsy
  • Plasma supports autoantibody immunoassays, multiplex cytokine panels, proteomics and metabolomics from a single acellular sample
