Allergy Leukopak for Type 2 Immunity and Allergic Disease Research

LeukoCore Allergy Leukopak biospecimens are collected from IRB-consented donors clinically diagnosed with allergy through IRB-approved apheresis procedures. Each leukopak contains large quantities of peripheral blood mononuclear cells, is processed and shipped within 24 hours of collection, and is available fresh or cryopreserved. Research Use Only (RUO).

Allergic Immunology and Why a Leukopak

  • Allergic disease is a type 2 immune response, not an autoimmune one: sensitization to an environmental or food allergen drives class switching to allergen-specific IgE rather than autoantibody production.
  • Allergen-specific IgE binds the high-affinity receptor FcεRI on mast cells and basophils; re-exposure crosslinks receptor-bound IgE and triggers immediate mediator release.
  • The adaptive arm is dominated by Th2-polarized CD4 T cells producing IL-4, IL-5, and IL-13, supported by epithelial alarmins and group 2 innate lymphoid cells; IL-5 drives eosinophil recruitment and survival.
  • These are the pathways targeted by approved biologics against IgE, IL-4 receptor alpha, and IL-5, so functional immune cells are central to mechanism and screening work.
  • A leukopak delivers far more cells per donor than a standard blood draw, which is what makes basophil activation testing, allergen-specific T cell expansion, and multi-assay panels from a single donor practical.
  • High per-donor yield also enables repeat sampling designs and reduces donor-to-donor variability across large screening campaigns.
