About Allergy PBMC

Allergy PBMC for IgE-Mediated Hypersensitivity and Tolerance Research

Allergy PBMC are sourced from IRB-consented donors clinically diagnosed with allergic disease, processed within 24 hours of collection and cryopreserved in CryoStor® CS10. Allergy is a type I, IgE-mediated hypersensitivity — a type 2 immune response to otherwise harmless environmental or dietary proteins — and is not an autoimmune disease. These cryopreserved peripheral blood mononuclear cells support the cellular immunology behind sensitisation, effector responses and the induction of tolerance.

Why PBMC for Allergic Disease

Serum tells you that a donor is sensitised. PBMC tell you why, and whether that sensitisation can be reversed — which is the central question in allergen immunotherapy and tolerance research.

  • Allergen-specific T cells are the upstream driver — allergen-reactive CD4+ Th2 cells produce the IL-4 and IL-13 that instruct B cells to class-switch to IgE. PBMC allow allergen-specific stimulation, proliferation and cytokine recall assays that no serum measurement can substitute for.
  • Type 2 cytokine readouts at the single-cell level — IL-4, IL-5 and IL-13 secretion measured by ELISpot, intracellular cytokine staining or cytokine capture requires viable cells and gives a functional rather than serological picture of sensitisation.
  • Regulatory mechanisms of tolerance — successful allergen immunotherapy is associated with induction of regulatory T cells, IL-10 production and a shift toward IgG4. PBMC are how those tolerance mechanisms are measured, making them central to immunotherapy mechanism-of-action work.
  • Biologic drug targets are cellular — anti-IgE and anti-IL-4 receptor alpha therapies act on cell-surface receptors and signalling, so receptor expression, occupancy and downstream signalling are assayed in intact cells.
  • Antigen-presenting cell biology — monocyte-derived dendritic cells from allergic donors support work on allergen uptake, processing and Th2-skewing capacity.
  • Complementary specimen note — basophil activation testing is normally performed on fresh whole blood rather than cryopreserved PBMC; contact us if you need a fresh cellular format for that application.

  • Sensitisation profile where reported: aeroallergen (pollen, dust mite, animal, mould), food, venom, or drug allergy
  • Total and allergen-specific IgE values where recorded, and skin prick test results where available
  • Atopic comorbidities: asthma, allergic rhinitis, chronic rhinosinusitis, atopic dermatitis
  • Immunotherapy status: immunotherapy-naive, or receiving subcutaneous, sublingual or oral allergen immunotherapy, with duration where recorded
  • Biologic therapy status: anti-IgE, anti-IL-4 receptor alpha, or biologic-naive
  • History of anaphylaxis and severity of reactions where documented
  • Matched healthy non-atopic controls available, age- and sex-matched on request
