Celiac Disease CD3+ T Cells are isolated from the peripheral blood of IRB-consented adult donors diagnosed with celiac disease, processed within 24 hours of collection and cryopreserved in CryoStor CS10. These enriched CD3+ T lymphocytes support antigen-specific T cell work in one of the best-characterised human autoimmune diseases.
Celiac Disease T Cell Biology and Why CD3+ Cells
Celiac disease is an autoimmune enteropathy triggered by dietary gluten in genetically susceptible individuals, and it is unusual in that both the HLA restriction and the driving antigen are known.
- Nearly all patients carry HLA-DQ2 or HLA-DQ8, which present deamidated gluten peptides to CD4+ T cells; HLA typing is effectively a prerequisite for antigen-specific experiments
- Tissue transglutaminase deamidates glutamine residues in gluten peptides, increasing their affinity for DQ2 and DQ8 and creating the immunodominant epitopes
- Gluten-specific CD4+ T cells provide help that drives autoantibody production against tissue transglutaminase, the basis of standard anti-tTG serology
- Intraepithelial cytotoxic lymphocytes mediate the villous atrophy and crypt hyperplasia seen on duodenal biopsy, with IL-15 implicated in their activation
- Gluten-specific CD4+ T cells are detectable in peripheral blood, particularly after gluten challenge, which is why a CD3-enriched peripheral product is a practical alternative to biopsy-derived cells
- CD3+ enrichment removes monocytes and B cells that confound T cell readouts, giving cleaner tetramer staining, proliferation assays and TCR repertoire sequencing
