Myasthenia Gravis PBMC for Autoimmune Neuromuscular Junction Research
Human Myasthenia Gravis PBMC products are sourced from IRB-consented donors clinically diagnosed with myasthenia gravis (MG) and processed under stringent conditions within 24 hours of collection. MG is an antibody-mediated autoimmune disease of the neuromuscular junction, driven by IgG autoantibodies against the acetylcholine receptor (AChR) or muscle-specific kinase (MuSK). These cryopreserved peripheral blood mononuclear cells give direct access to the autoreactive B-cell and T-cell compartments that generate and sustain those antibodies.
Disease Biology and Why PBMC
- Autoreactive B cells and plasmablasts are the source of pathogenic AChR and MuSK autoantibodies; PBMCs allow their identification, sorting, and single-cell characterisation.
- Antigen-specific T-cell help: AChR-reactive CD4+ T cells and follicular helper T cells drive class-switched autoantibody production and can be profiled by tetramer, activation-induced marker, or cytokine assays.
- Regulatory T-cell function is impaired in MG and is a recognised research focus for tolerance-restoring therapy.
- MuSK-positive disease is IgG4-predominant, giving a distinct B-cell biology from complement-driven AChR-positive disease and a rationale for stratified cellular studies.
- B-cell-depleting and CAAR-T approaches: PBMCs support target engagement, repopulation, and cytotoxicity work for rituximab, anti-CD19/CD20, BAFF-directed, and chimeric autoantibody receptor T-cell programmes.
- Thymic biology: thymoma-associated and hyperplastic donors provide a peripheral window on a disease with a strong thymic component.
