Human Peanut Allergy Serum for IgE and Allergen Component Research
Our Peanut Allergy Serum is sourced from IRB-consented donors clinically diagnosed with peanut allergy. Each sample is processed and frozen within 24 hours of collection to ensure consistency and high quality for allergy immunology and diagnostic development. Peanut allergy is a type I, IgE-mediated hypersensitivity in which allergen-specific IgE bound to FcepsilonRI on mast cells and basophils is cross-linked by peanut protein, producing immediate degranulation — an allergic mechanism, not an autoimmune one.
Why Serum for Peanut Allergy Research
- Serum is the reference matrix for quantitative allergen-specific IgE. The clinical assays and the published thresholds relating specific IgE concentration to reaction probability were established in serum, making it the correct choice for comparability.
- Component-resolved diagnostics rests on serum: sensitisation to the individual peanut storage proteins Ara h 1, Ara h 2, Ara h 3 and Ara h 6 carries different clinical weight, with Ara h 2 the most clinically informative single component.
- Allergen-specific IgG4 rises during oral immunotherapy, and the balance between specific IgE and specific IgG4 is one of the best-characterised serological correlates of desensitisation. Both isotypes are measured in serum.
- Serum from allergic donors serves as the IgE source for indirect basophil activation and passive sensitisation assays, where donor serum is used to sensitise reporter cells from a non-allergic donor.
- Cross-reactivity between peanut and tree nut or legume allergens is a serological question, addressed by inhibition and component profiling in serum rather than by cellular assay.
- Because serum contains no anticoagulant, it avoids citrate or EDTA interference on the immunoassay and mass spectrometry platforms most commonly used in allergy diagnostics.
