Systemic Lupus Erythematosus (SLE) Plasma is collected from IRB-consented donors clinically diagnosed with SLE and processed within hours of collection to preserve cytokines, complement components, circulating nucleic acids, and metabolites. This human plasma supports research into immune complex biology, interferon signalling, and biomarker discovery in SLE. Bulk plasma is available.
Why Plasma for Systemic Lupus Erythematosus Research
SLE is a systemic autoimmune disease in which tolerance to nucleic acid–containing self antigens is lost. Its pathology is largely mediated by circulating immune complexes and the interferon response they trigger, which makes the fluid phase of blood the site of action. Plasma is collected with anticoagulant and separated without a clotting step, which is important for complement fragments and cell-free nucleic acids that clotting perturbs.
- Immune complexes are the pathogenic unit — antibodies against double-stranded DNA, Smith antigen, and other ribonucleoproteins form complexes that deposit in tissue and activate complement; plasma is where they circulate
- Complement consumption is a direct disease readout — falling C3 and C4 with rising activation fragments reflects active complement turnover, and anticoagulated plasma is the preferred matrix for complement fragment work
- Type I interferon signature — nucleic acid–containing complexes engage TLR7 and TLR9 in plasmacytoid dendritic cells, driving the interferon response that anti-interferon receptor therapy targets; interferon-inducible proteins are measurable in plasma
- Cell-free DNA and nucleosomes — impaired clearance of apoptotic material leaves circulating nucleosomes and cell-free DNA that are both antigen source and candidate biomarker, and require unclotted plasma to measure reliably
- B cell survival signalling — BAFF is elevated in SLE and is the target of established B cell–directed therapy, making it a natural plasma analyte for stratification
- Activity is dynamic — SLE alternates between flare and quiescence, so serial or activity-defined sampling is far more informative than a single unclassified draw

- Renal involvement — biopsy-proven or clinically diagnosed lupus nephritis versus non-renal SLE
- Organ involvement — cutaneous, articular, haematological, or neuropsychiatric manifestations where documented
- Treatment status — hydroxychloroquine, corticosteroids, mycophenolate or other immunosuppressant, or biologic therapy including B cell– and interferon-directed agents
- Matched healthy controls available — demographically matched healthy donor plasma
