Type 1 Diabetes Plasma for Autoimmune Beta-Cell Destruction Research

Type 1 Diabetes Plasma is collected from IRB-consented donors clinically diagnosed with type 1 diabetes (T1D) and processed within one day of collection to preserve key analytes such as cytokines, proteins, and metabolites. Type 1 diabetes is an autoimmune disease: autoreactive CD8+ T cells destroy insulin-producing pancreatic beta cells, and islet autoantibodies against GAD65, IA-2, ZnT8 and insulin mark the underlying loss of tolerance. This is a mechanistically distinct condition from the insulin resistance of type 2 diabetes. Bulk plasma is available.

Why Plasma for Type 1 Diabetes Research

• C-peptide is the accepted measure of residual endogenous insulin secretion and therefore of surviving beta-cell mass. It is the primary endpoint in most beta-cell preservation trials and is readily quantified in plasma.
• Beta-cell death releases beta-cell-derived cell-free DNA into the circulation, detectable by differentially methylated insulin gene sequences. Cell-free DNA work requires plasma rather than serum, because clotting releases leukocyte genomic DNA that dilutes the signal of interest.
• Plasma supports the metabolic and inflammatory readouts that accompany autoimmune beta-cell loss, including cytokine and chemokine panels and markers of the innate response implicated in islet inflammation.
• Glucose is more stable in appropriately anticoagulated plasma than in serum left in contact with cells, which matters for any study relating glycaemia to immune or metabolic analytes in the same draw.
• Because type 1 diabetes progresses through defined preclinical stages, plasma is well suited to longitudinal designs in which the same donor supplies serial samples across changing residual beta-cell function.
• A note on matrix choice: islet autoantibody testing for GAD65, IA-2, ZnT8 and insulin is conventionally performed in serum, where the reference standardisation sits. Use the serum product for autoantibody work and plasma for C-peptide, cell-free DNA and omics studies.
