Rheumatoid Arthritis Human PBMC products are sourced from IRB-consented donors clinically diagnosed with rheumatoid arthritis (RA), processed within 24 hours of collection and cryopreserved in CryoStor CS10. These peripheral blood mononuclear cells support T cell, B cell and monocyte studies relevant to autoimmune activity and therapeutic target validation.
RA Autoimmunity and Why PBMC
Rheumatoid arthritis is a systemic autoimmune disease that targets synovial joints, producing chronic synovitis and, if uncontrolled, progressive cartilage and bone erosion.
- Anti-citrullinated protein antibodies (ACPA) and rheumatoid factor define the seropositive subset, and ACPA can precede clinical arthritis by years
- The shared epitope alleles of HLA-DRB1 are the dominant genetic risk factor, and PTPN22 variants contribute additional risk
- CD4+ T cell help, including Th17 responses, drives B cell activation and autoantibody class switching; B cell depletion is an effective clinical strategy, which underlines the B cell contribution
- Monocytes and macrophages are the principal sources of TNF-alpha and IL-6, the cytokines targeted by the most widely used biologic therapies
- Peripheral monocyte, T cell and B cell phenotypes shift with disease activity and with treatment, making viable PBMC the appropriate specimen for mechanism and pharmacodynamic work
- PBMC support high-parameter flow cytometry, single-cell sequencing, JAK-STAT and cytokine signalling assays, and ex vivo drug response testing
