HIV CD19+ B Cells for Viral Immunology and Antibody Discovery
HIV CD19+ B Cells are purified from peripheral blood of IRB-consented adult donors diagnosed with Human Immunodeficiency Virus (HIV). These B lymphocytes carry the characteristic B cell abnormalities of chronic HIV infection and support studies of humoral immune dysfunction, vaccine responsiveness, and antibody discovery. Cryopreserved for optimal viability.
What HIV Does to the B Cell Compartment
Although HIV infects CD4+ T cells rather than B cells directly, the B cell compartment is substantially disturbed — and in ways that make donor material genuinely valuable rather than merely disease-labelled:
- Polyclonal hypergammaglobulinemia — elevated total immunoglobulin from chronic activation, alongside paradoxically poor antigen-specific responses
- Exhausted “tissue-like memory” B cells — a CD21-low, inhibitory-receptor-high population expanded in viremic donors
- Loss of resting memory B cells, which underlies impaired responses to vaccination even in donors on suppressive therapy
- Increased apoptosis and altered BCR signalling
- Broadly neutralizing antibody (bNAb) development — a subset of donors generates bNAbs after years of infection, and their B cells are the source material for isolating and characterising those lineages
That last point is why HIV donor B cells matter well beyond HIV itself: bNAb lineage tracing from elite neutralisers underpins much of current HIV vaccine immunogen design.
